چکیده

Our long term goal is to determine the role that pMGA hemagglutinins play in Mycoplasma gallisepticum F-strain (termed as F-strain for brevity) persistence in poultry. To do this however, we first had to identify the pMGA genes expressed by an isolate of the F-strain. To identify F-strain pMGA genes, a genomic library was constructed in bacteriophage lambda and screened with polyclonal antisera raised against F-strain surface antigens capable of inhibiting agglutination of chicken red blood cells. One phage clone was identified, designated 8 9, which remained positive through additional rounds of screening and was sequenced. Sequence data analysis predicted one complete and two partial open reading frames (ORF) arranged in tandem within the 4.5 kb genomic insert, termed ORF 9.1, ORF 9.2 and ORF 9.3, which showed significant homology to the pMGA gene family of M. gallisepticum. The number of GAA trinucleotide repeats in the intergenic region of 9.3 ORF suggested that this ORF is expressed. The pMGA cDNAs amplified from the F-strain showed significant homology to the 9.3 ORF. One set of cDNAs however, differed from the 9.3 ORF sequence by a repeat of the sequence “AACCAAA” in the 5' end. We conclude that the F-strain vaccine isolate expresses pMGA gene variants, similar to what has been described in field strains of M. gallisepticum.

Molecular characterization of Mycoplasma synoviae isolated from broiler chickens of West Azarbaijan province by PCR of vlhA gene

Mycoplasma synoviae (MS) is a pathogen responsible for respiratory and locomotor disorders and causes major economic losses in poultry industry. Early and accurate diagnosis of MS infection plays a major role in control of the infection. This study was conducted to characterize Iranian field isolates of MS isolated from broiler chickens of West Azarbaijan province (Northwest of Iran), ...

S1 gene sequence analysis of infectious bronchitis virus vaccinal strains (H120 & H52) and their embryo-passaged derivatives

Avian infectious bronchitis is an acute and highly contagious disease that mainly causes respiratory symptoms in poultry. A number of serotypes and variants of the viral agent with poor cross-protection are the major problem to achieve desired immunity from vaccination. The S1 subunit of S glycoprotein (spike) is the major determinant of IBV so that a minor change in amino acid sequence of this...

Molecular characterization of Mycoplasma synoviae isolates from commercial chickens in Iran

Detection of Mycoplasma synoviae (MS) by culture and polymerase chain reaction (PCR) has been reported from commercial chicken farms in different provinces of Iran. In some reports the phylogenetic analysis of MS isolates based on 16S rRNA and variable lipoprotein hemagglutinin (vlhA) genes have been carried out. The PCR product containing partial 16S rRNA genes of Iranain isolates was sequence...

Size and genomic location of the pMGA multigene family of Mycoplasma gallisepticum.

The pMGA multigene family encodes variant copies of the cell surface haemagglutinin of Mycoplasma gallisepticum. Quantitative Southern blotting, using an oligonucleotide probe complementary to a region conserved in the leader sequence of all known pMGA genes, was used to estimate the number of members of the family in the genome of seven strains of M. gallisepticum. The number of copies estimat...
